| BIOLOGicalSignificance | TightjunctionsfunctiontomaintaincellularpolarityandpermeABIlitybarriersinepithelialandendothelialcells.Recently,severalcomponentmoleculesoftightjunctionhavebeenidentified.The7H6antigenisconsideredanimportantmoleculeforregulationofthebarrierfunctionoftightjunctionsfromexaminationofMADInDarbycaninekidney(MDCK)cells.Expressionofthe7H6antigenwasalsoexaminedduringrathepatocarcinogenesisandinhumancancertissues.Semiquantitativeanalysisbyconfocallaserscanningmicroscopyrevealedthattheexpressionof7H6antigendecreasedsequentiallyinpreneoplasticlesionsandcancertissuesappearedintheliverduringhepatocarcinogenesis.The7H6antigenwasexpressedscarcelyinHCCwithatrabecularpattern,whereasitwasexpressedintenselyattheapicalandbasolateralmembraneofcancercellswithglandularpattern.Examinationofhumangastricandcoloncancertissuesshowedthattightjunctionsweremaintainedhighlyinthewell-differentiated(gland-forming)adenocarcinomas,butreducedinexpressioninthepoorly-differentiatedadenocarcinomas.Theseindicatethatimpairmentoftightjunctionsmaybeassociatedwithmalignantphenotypesofcancercells,suchasthelossofcellularpolarityandadhesiveness. |
