| BIOLOGicalSignificance | The6B11monoclonalantibodyreactswithanepitopeoftheCDR3formedbythegerm-lineconfigurationoftheValpha24andJalpha18oftheTCRalphalocus,whichresultsintheexpressionofaninvariantT-cellreceptorexpressedbyinvariantNKTcells.ThisTCRishomologoustothemurineValpha14Jalpha18.ThisTCRalphachainpairswitharestrictedsetofrearrangedTCRbetachains,withVbeta11beingthemostprominentinhumans.NKTcellsarerestrictedbytheCD1dmoleculeandareactivatedbyaCD1d-presentedglycolipidligand,whichresultsintherapidproductionofIL-4,IL-13andIFNgamma.AlthoughiNKTcellshaveNK-likecytolyticactivity,theyareconsideredregulatorsofimmuneresponsesbecausetheyrapidlyproducelargeamountsofbothTh1andTh2cytokinesinautoimmunedisease,infectiousdisease,andcancer.ThenumberofiNKTcellspresentinperipheralbloodofnormalhumansshowsconsiderablevariabilityrangingfrom0.01-1%oftotalperipheralTcells. |
