| BIOLOGicalSignificance | TheTB2FantibodyreactswithTransformingGrowthFactor-beta,apleiotropiccytokine,existsinfiveisoforms,knownasTGF-beta1-5.Homologiesbetweenisoformsrangefrom70-80%butnohomologyexiststoTGF-alpha.TGF-beta1isubiquitousandthemostabundantformfoundinlymphoidorgans,whileotherisoformsareexpressedinamorerestricteddistribution.Thebiologicallyactivestateofallisoformsaredisulfide-linkedhomodimers.Theheat-andacid-stablemonomericsubunitshavealengthof112aminoacids.TheisoformsofTGF-betaarisebyproteolyticcleavageoflongerprecursorstheisoformsarederivedfromthecarboxyterminalendsoftheseprecursors.Isoformsisolatedfromdifferentspeciesareevolutionarilycloselyconservedandhavesequenceidentitiesontheorderof98%.Maturehuman,porcine,simian,chickenandbovineTGF-beta1areidenticalanddifferfrommouseTGF-beta1inasingleaminoacid.TGF-beta1isproducedinveryhighlevelsbyplatelets.OthercellularsourcesofTGF-beta1includemacrophages,lymphocytes,endothelialcells,chondrocytes,andleukemiccells.TGF-beta1secretioncanbeinducedbysteroids,retinoids,EGF,NGF,vitaminD3,andIL-1.ActivitiesofTGF-beta1includeinhibitionofcellgrowthforinhibitorfornormalandtransformedepithelialcells,endothelialcells,fibroblasts,neurons,andlymphoidcellsandotherhematopoieticcelltypes.TGF-beta1inhibitstheproliferationofTcellsandNKcellsanddownregulatestheactivitiesofactivatedmacrophages.TGF-beta1blockstheanti-tumoractivityofIL-2?bearinglymphokine-activatedkillercells.Recently,TGF-beta1hasbeenfoundtohaveacriticalroleinthedevelopmentofregulatoryTcells.DendriticcellsexposedtotumorshavebeenreportedtosecreteTGF-beta1andstimulateexpansionofnaturalTregcells.Moreover,TGF-beta1hasbeenshowntoactasacostimulatoryfactorforexpressionofFoxp3,leADIngtothedifferentiationofCD4+CD25+TregcellsfromperipheralCD4+CD25-Progeny.TGF-beta-inducedregulatoryTcellshavebeentermedTi-Treg. |
