| Biological Significance | CD14 cooperates with MD-2 and TLR4 to mediate the innate immune response to bacterial lipopolysaccharide (LPS). CD14 acts via MyD88, TIRAP and TRAF6, leading to NF-kappa-B activation, cytokine secretion and the inflammatory response. CD14 also up-regulates cell surface molecules, including adhesion molecules. |
| Expression | CD14 is preferentially expressed on monocytes/macrophages.CD14 is also expressed by several non-myeloid cells. CD14 is expressed mainly by macrophages and (at 10-times lesser extent) by neutrophils. It is also expressed by dendritic cells. The soluble form of the receptor (sCD14) is secreted by the liver and monocytes and is sufficient in low concentrations to confer LPS-responsiveness to cells not expressing CD14. mCD14 and sCD14 are also present on enterocytes. It acts via MyD88, TIRAP and TRAF6, leading to NF-kappa-B activation, cytokine secretion and the inflammatory response, up-regulates cell surface molecules, including adhesion molecules. sCD14 is also present in human milk, where it is believed to regulate microbial growth in the infant gut. |
