| BIOLOGicalSignificance | HeparincofactorII(HCII),asinglechainglycoproteinwithaMWof65kDa,isaserineproteaseinhibitorthatissynthesizedbytheliverandcirculatesinplasma.HeparincofactorIIinhibitsthrombinbyformationofastablebimolecularcomplex,buthasnoactivityagainstotherproteasesinvolvedincoagulationorfibrinolysis.TherateatwhichHCIIinhibitsthrombinincreasesmorethan1000-foldinthepresenceofheparin,heparansulfate,ordermatansulfate.HeparincofactorIIisuniqueamongserineproteaseinhibitorsinitsABIlitytobestimulatedbydermatansulfate,anditbindstoaminorsubpopulationofdermatansulfateoligosaccharides.TurnoverstudiesoflabeledHCIIinhumanssuggestthat40%oftheproteinequilibrateswithanextravascularcompartment,butthedistributionofHCIIinvarioustissueshasnotbeenthoroughlyinvestigated.HeparincofactorIIhasbeendetectedintheintimaofnormalhumanarteries,andtheabilityofdermatansulfateinthearterialwalltostimulateHCIIisdecreasedinatheroscleroticlesions. |
